Abstract

¸ñÀû:Ä¡ÁÖÁ¶Á÷Àç»ýÀ» À§Çؼ­´Â Ä¡ÁÖÀδ뼼Æ÷ÀÇ Áõ½Ä ¹× ÀÌÁÖ¸¦ ÃËÁø½ÃÅ°´Â °ÍÀÌ ÇʼöÀûÀ̳ª ÇöÀç±îÁö ÀÌ°ÍÀ» ¸¸Á·½ÃÅ°´Â Àç»ý¼ú½ÄÀº ¾ø´Ù. Àç·á ¹× ¹æ¹ý: °Ç°­ÇÑ ¼ºÀÎÀÇ Á¦ 3 ´ë±¸Ä¡·ÎºÎÅÍ Ä¡ÁÖÀδ뼼Æ÷¸¦ ÃßÃâÇÑ ÈÄ, ¹ý¶û±âÁúÀ¯µµÃ¼(Emdogain (Biora, Malmo, Sweden))ÀÇ ³óµµ°¡ °¢°¢ 0, 12.5, 25, 50, 100, and 200 ug/mL ÀÎ ¹èÁö¿¡¼­ ¹è¾ç½ÃÄ×´Ù. Ä¡ÁÖÀÎ´ë ¼¼Æ÷Áõ½Ä°ú ¾ËÄ®¸®¼º ÀλêºÐÇØÈ¿¼Ò(alkaline phosphatase) È°¼ºµµ¸¦ ÃøÁ¤ÇÏ°í, ¹ßÇöµÇ´Â ¼ºÀåÀÎÀÚ¸¦ Æò°¡ÇÏ¿´´Ù.

°á·Ð: ¹ý¶û±âÁú À¯µµÃ¼´Â Àΰ£ Ä¡ÁÖÀδ뼼Æ÷ÀÇ ¼¼Æ÷Áõ½Ä°ú ¾ËÄ®¸®¼º ÀλêºÐÇØÈ¿¼Ò È°¼ºÀ» ÁõÁø½ÃÅ°°í, VEGF¿Í TGF-B µî ¼ºÀåÀÎÀÚÀÇ ¹ßÇöÀ» ÃËÁøÇÔÀ¸·Î½á Ä¡ÁÖÁ¶Á÷ Àç»ý¿¡ ±â¿©ÇÒ ¼ö ÀÖÀ» °ÍÀÌ´Ù.

PURPOSE. Stimulating the proliferation and migration of periodontal ligament cells (PDLCs) has become the main goal of periodontal regeneration. To accomplish this goal, regeneration procedures have been developed, but results have not been predictable. Recently, tissue engineering using enamel matrix derivatives (EMDs) and growth factors has been applied to periodontal regeneration; however, the mechanism of EMDs is largely unknown. The aim of this study was to investigate the effects of EMDs on the proliferation and release of growth factors from PDLCs.

MATERIALS AND METHODS. Human PDLCs were removed from individually extracted 3rd molars of healthy young adults, and cultured in the media containing EMDs (Emdogain, Biora, Malmo, Sweden) at concentration of 0, 12.5, 25, 50, 100, and 200 ug/mL each. Cell proliferation and ALP (alkaline phosphatase) activity were measured. The evaluation of growth factors released by PDLCs was also performed by one-way analysis of variance (ANOVA) and Bonferroni¡¯s multiple comparison test.

RESULTS. Significantly increased proliferation and ALP activity were observed in PDLCs treated with over 25 ug/mL and 50 ug/mL EMDs, respectively. Additionally, treatment of PDLCs with 50 ug/mL resulted in significantly increased release of vascular endothelial growth factor (VEGF) and transforming growth factor (TGF)-B after 24 h and 48 h, respectively.

CONCLUSION. EMDs enhance the proliferation and ALP activity of PDLCs, and promote the release of growth factors, including VEGF and TGF-B, from PDLCs. Therefore EMDs could be one of the effective methods for periodontal regeneration.
